      目的: 利用芯片技术探讨黄秦艽对雪上一枝蒿所致小鼠心脏急性毒性基因表达的影响及分子机制.方法: 25 只雄性昆明小鼠随机分为正常对照组、 雪上一枝蒿氯仿部位组(CFA) 和黄秦艽水提物组(WVBF) , 通过表达谱 对各组心脏差异表达基因进行生物信息学研究. 结果: WVBF对CFA诱导的急性心脏毒性具有明显的解毒效果, 染毒组与正常组相比,差异基因的功能主要涉及机体内物质的异常代谢,WVBF主要通过干扰CFA引起的内质网 应激反应及线粒体功能紊乱过程,提高细胞存活率,使心肌恢复正常功能.结论: 黄秦艽可以通过多条信号通路缓 解雪上一枝蒿所致的小鼠心肌毒性作用.
英文摘要:
      Aim: To investigate the effect and mechanism of Veratrilla Baillonii on the acute cardiac toxicity and on gene expression in mice treated with Aconitum Brachypodum by using gene chip technique. Methods: 25 male mice were divided randomly into control group,chloroform fraction of Aconitum Brachypodum Diels (CFA) group and water extract of Veratrilla Baillonii (WVBF) group. Take mice hearts to study the expressions of various genes for bioinformatics research.Results: The results showed that WVBF had a significant detoxification effect on CFA-induced acute cardiac toxicity. Gene Ontology (GO) analysis mainly related to abnormal metabolism of the body. Pathway analysis showed that WVBF modulated the process of endoplasmic reticulum stress and mitochondrial dysfunction induced by CFA, thus improve cell survival rates and restore normal function to myocardium. Conclusions: WVBF could attenuate CFA-induced cardiac damage in mice through multiple signaling pathways.
