      目的:探究黄精多糖对环磷酰胺诱导小鼠免疫抑制的拮抗作用.方法:采用MTT法体外评价了黄精多糖对正常小鼠脾细胞的增殖活性,用环磷酰胺制备了小鼠体免疫抑制模型,并分别测定了小鼠脾脏、胸腺指数,脾脏、胸腺细胞的体外增殖活力,巨噬细胞吞噬功能,巨噬细胞分泌炎性细胞因子的能力. 结果:黄精多糖能显著提高正常小鼠脾细胞增殖活力,当剂量大于200 μg·mL-1时,协同ConA增殖促进率在700%以上;黄精多糖能提高免疫功能低下小鼠的脾脏和胸腺指数,促进ConA刺激下的脾脏和胸腺细胞增殖,增强巨噬细胞的吞噬功能和IL-6和TNF-α的分泌.与模型组比较,高剂量黄精多糖给药组(150 mg·kg-1)小鼠脾脏指数增加了79.0%,协同ConA刺激指数增加了43.3%;胸腺指数升高了148.3%,协同ConA刺激指数升高48.5%.结论:黄精多糖能显著缓解环磷酰胺所致的小鼠免疫抑制.

      Objective: To investigate the effects of polygonatum sibiricum polysaccharide(PSP) on the immunosuppression activities induced by cyclophosphamide in mice. Methods: MTT assay was used to evaluate the proliferation activity of splenocytes of normal mice. Mice immunosuppression model was established with cyclophosphamide. The mice spleen, thymus index and spleen cell, thymocyte proliferation activity in vitro, macrophage phagocytosis, and the ability of macrophages to secrete inflammatory cytokines were respectively determined. Result: PSP could evidently increase the proliferation activity of spleen cell of normal mice. The synergistic proliferation promotion rate with ConA was more than 700% with PSP dose of 200 μg·mL-1. PSP could also improve the spleen and thymus index of immunosuppressed mice, promote the proliferation of spleen and thymus cells stimulated by ConA, and enhance the phagocytosis of macrophage and the secretion of IL-6 and TNF-α. Compared with the control group, the spleen index increased by 79.0%, 43.3% in high PSP dose (150 mg·kg-1) for ConA synergized group, and the thymus indexes increased by 148.3%, 48.5% respectively. Conclusion: PSP could significantly alleviate the immunosuppression of mice induced by cyclophosphamide
