      目的:探究龙血竭阻断Kv1.3通道的主要活性组分.方法:龙血竭总黄酮经优化工艺提取后,采用全细胞膜片钳实验检测其对内源性和外源性表达的Kv1.3通道调节作用.结果:龙血竭总黄酮对内源性和外源性表达的Kv1.3通道均具有较强的抑制作用,呈浓度依赖性和通道选择性.结论:总黄酮介导了龙血竭阻断Kv1.3通道,为进一步发现龙血竭中Kv1.3通道阻断剂提供了重要线索.
英文摘要:
      Objective: To elucidate the main active components of Resina Draconis in blocking Kv1.3 channels. Methods: The total flavonoids from Resina Draconis (tFRD) were extracted with the optimized extraction process, and their effect on endogenous and exogenous Kv1.3 channels were detected by whole cell patch clamp experiments. Results: The tFRD showed strong inhibitiory effect on endogenous and exogenous Kv1.3 channels in a concentration-dependent and channel selective manner. Conclusions: Total flavonoids mediated the blocking effect of Resina Draconis in Kv1.3 channels, which provided important clues for discovery of further Kv1.3 channel blockers from Resina Draconis .
