      目的:探讨垂盆草乙醇提取物(EESS)对HEK293T细胞凋亡的诱导作用和机制.方法:利用CCK-8法检测垂盆草乙醇提取物对细胞增殖影响,用Hoechst33342染色法及Annexin V-FITC染色流式细胞检测其对细胞凋亡的影响,以Western blot检测其对NF-κB信号通路蛋白及凋亡相关通路蛋白Bcl-2, Bax, Bcl-xL等表达的影响. 结果:EESS对细胞增殖的抑制并促进其凋亡呈剂量依赖性,能显著上调NF-κΒ磷酸化水平及凋亡启动蛋白线粒体细胞色素C(cytochrome C, Cyc)蛋白和促凋亡蛋白Bax的表达(P<0.05).结论:垂盆草乙醇提取物能诱导HEK 293T细胞的凋亡,其作用机制与NF-κΒ信号途径及其介导的凋亡启动蛋白Cyt c和促凋亡蛋白Bax的表达上调有关.

      Objective: To investigate the effect and mechanism of ethanol extract of Sedum sarmentosum Bunge (EESS) on inducing HEK293T cells apoptosis. Method: The effect of EESS on cell proliferation was detected by CCK-8 method, and its effect on cell apoptosis was detected by Hoechst33342 staining and Annexin V-FITC staining flow cytometry. The impacts of EESS on the expression of NF-κB signaling pathway proteins and apoptosis-related proteins, Bcl-2, Bax and Bcl-xl were determined by Western blot. Results: EESS inhibited cell proliferation and promoted apoptosis in a dose-dependent manner. Furthermore, it could significantly up-regulate the NF-κΒ phosphorylation level and the expression of Cyc protein and pro-apoptotic protein Bax (P<0.05). Conclusion: EESS can induce the apoptosis of HEK 293T, and the mechanism of which is related to the up-regulation of NF-κΒ signaling pathway and its expression of the apoptosis-inducing protein Cyc and the pro-apoptotic protein Bax.
