      目的:以网络药理学知识为基础,探寻忧遁草治疗乙型肝炎的活性成分与作用机制. 方法:基于传统中草药数据库与化合物信息数据库全面挖掘忧遁草活性成分,利用虚拟对接平台预测筛选各组分靶蛋白,通过化合物-蛋白-疾病互作网络确定忧遁草治疗慢性乙型肝炎的重要靶点,结合生物信息学手段、GO分析及KEGG信号通路富集,分析忧遁草治疗慢性乙型肝炎的活性成分及分子机制. 结果: 获得忧遁草类药成分27种,其中治疗乙型肝炎有效成分16种,数据库挖掘及维恩分析获得疾病-化合物重叠靶基因20个,筛选出关键节点与Hub节点,富集分析显示忧遁草可能通过凋亡、炎症、免疫等多个方面,对多个靶点、多个通路产生调控作用. 结论:基于网络药理学研究表明,忧遁草可能通过多重作用靶点,参与调控多条主要信号通路,影响多类细胞的功能,从而起到治疗乙型肝炎的作用,为阐释其治疗乙型肝炎的物质基础与作用机制提供了科学依据.
英文摘要:
      Objective: Based on the knowledge of network pharmacology, explore the active ingredients and mechanism of action of clinacanthus nutans in treating hepatitis B. Methods: Based on the traditional Chinese herbal medicine database and compound information database, the active components of clinacanthus nutans were fully explored. Using the virtual docking platform to predict and screen each component target protein, the compound-protein-disease interaction network is used to determine the important target of the treatment of chronic hepatitis B by Clinacanthus nutans, combined with bioinformatics, GO analysis and KEGG signaling pathway enrichment. The active ingredients and molecular mechanism of Clinacanthus nutans were analyzed in treating chronic hepatitis B. Results: Twenty-seven kinds of medicinal herbs were obtained, among which 16 kinds of effective components of hepatitis B were treated. Database mining and venn analysis obtained 20 disease-compound overlapping target genes.The enrichment analysis showed that Clinacanthus nutans could regulate multiple targets and multiple pathways through apoptosis, inflammation and immunity. Conclusion: Based on network pharmacology research, it is suggested that Clinacanthus nutans may participate in the regulation of multiple major signaling pathways through multiple targets, affecting the function of many types of cells, and thus play a role in the treatment of hepatitis B, in order to explain its therapeutic type B. The material basis and mechanism of action of hepatitis provide a scientific basis.
