      为确定复方中药榆芩颗粒的最佳提取工艺,采L9(34)正交试验进行优选,以干浸膏收率、没食子酸、鞣花酸和黄芩苷含量作为指标,考察了药材粒度、乙醇浓度、加醇量以及提取时间的影响,以此确定最佳提取工艺. 采用高效液相色谱法测定样品中没食子酸、鞣花酸和黄芩苷含量,采用C18为色谱柱(柱温保持30 ℃),乙腈-0.2%磷酸缓冲盐溶液为流动相,梯度洗脱,检测波长270 nm,这三种成分在各自范围内呈现良好的线性关系. 优选的最佳提取工艺为:石榴皮单提,药材粉碎为粗粉,乙醇浓度60%,第一次6倍量提取15 min,第二次4倍量提取10 min;其他药材粉碎为粗粉,乙醇浓度60%,第一次6倍量提取1 h,第二次4倍量提取0.5 h.

      In order to determine the best extraction technology of Yuqin granules, L9 (34) orthogonal test was used to optimize the extraction technology. The yield of dry extract, and the contents of gallic acid, ellagic acid and baicalin were used as evalution index to investigate the effects of powder size, ethanol concentration, alcohol dosage and extraction time. The contents of gallic acid, ellagic acid and baicalin were determined by High performance liquid chromatography (HPLC), HPLC was performed on the column of C18, the mobile phases was made of acetonitrile-0.2% phosphate buffer solution and the method of gradient elution was used, the detection temperature was 30 °C, the detection wavelength was 270 nm. These three indicator components showed good linear relationship in their respective ranges. The optimal extraction technology was as follows: Punica granatum L. was extracted separately and crushed into coarse powder, and then extracted with 60% ethanol. The first time was 15 min with 6 times volume, and the second extraction time was 10 min with 4 times volume. Sanguisorba officinalis L and Scutellaria baicalensis Georgi were crushed into coarse powder and extracted with 60% ethanol, and then extracted for 1 h with 6 times volume in the first time, and 0.5 h with 4 times volume in the second time.
