      为了研究白玉蜗牛中糖胺聚糖(AFG)的纯化、理化性质和化学结构,采用酶解-碱解提取白玉蜗牛粗多糖,以季铵盐沉淀法和离子交换柱层析法纯化获得AFG,得率为0.88%. 理化性质分析显示,AFG为均一性的酸性粘多糖(Mw为23,840 Da);单糖组成分析及1H NMR谱图显示,AFG是由等摩尔的D-乙酰氨基葡萄糖(D-N-acetyl glucosamine,GlcNAc)和L-艾杜糖醛酸硫酸酯(2-sulfated L-iduronic acid,IdoA2S)组成的糖胺聚糖;采用β-消除解聚法获得其解聚产物(dAFG),UV及1H/13C NMR显示其非还原端为不饱和己糖醛酸,与β-消除解聚法的糖苷键选择性解聚特点相符. 结论:白玉蜗牛所含AFG由等摩尔的GlcNAc和IdoA2S组成,其具有类似于肝素的结构特点;β-消除解聚可用于制备低分子量的AFG解聚产物.
英文摘要:
      o study the purification, physicochemical properties and chemical structure of glycosamino-glycan (AFG) from white jade snail(Achatina fulica), the crude polysaccharide of white jade snail was extracted by enzymolysis and then alkaline hydrolysis. AFG was purified by treatment with benzethonium chloride and anion exchange chromatography, and the yield is 0.88% .AFG is a homogeneous acidic mucopolysaccharide with Mw of 23 840 Da, andchemical composition analysis, IR and NMR spectra showed AFG is composed of 2-sulfated iduronic acid (IdoA2S) and N-acetyl-glucosamine (GlcNAc). AFG was depolymerized by chemical β-eliminative depolymerization method to yield dAFG.. The UV and 1H/13C NMR spectra showed that the structure of dAFG was consistent with AFG except the unsaturated uronic acid at the non-reducing end. Conclusion:AFG isolated from white jade snail (Achatina fulica)is composed of a uniform repeating disaccharide of 4-L-IodA2S-α1,4-D-GlcNAc-α1.
