      目的:为研究白茅根乙酸乙酯部提取物(EAERI)对小鼠气管平滑肌的舒张作用.方法:使用常用的激动剂(80 mmol.L-1 KCl或100 μmol.L-1 ACh)预刺激离体小鼠的气管环,进行张力检测和单细胞离子通道电流的记录.结果:EAERI可剂量依赖性地抑制由高钾和ACh诱导的气管环预收缩,而对静息状态下的气管无影响.EAERI还可以抑制L型钙通道(VDCCs)电流,阻断细胞外Ca2+内流. 其在抑制ACh诱导的收缩反应中除VDCCs通道外,还有其它离子通道,从而抑制肌质网内Ca2+释放,最终舒张小鼠气管平滑肌.结论:EAERI中的有效成分可以作为一种气管舒张剂,用于缓解哮喘.

      Objective: To investigate the relaxation effects of ethyl acetate extracts from Rhizoma imperate (EAERI) on mouse trachea smooth muscle. Methods: The tracheal loops of isolated mice were prestimulated with the usual agonists (80 mmol.L-1 KCl or 100 μmol.L-1 ACh) for tension detection and recording of single-cell ion channel currents. Results: EAERI dose-dependently inhibited preconstriction of the tracheal ring induced by high potassium and ACh but did not affect the tracheal in resting state. EAERI also inhibited L-type calcium channel (VDCCs) currents and blocked extracellular Ca2+ influx. In addition to the VDCCs channel, there might be other ion channels in the inhibition of ACh induced contraction response, thus inhibiting Ca2+ release in the sarcoplasmic reticulum, and finally relaxing the tracheal smooth muscle of mice. Conclusion: The active ingredient in EAERI can be used as a tracheal dilator to relieve asthma.
