      脱落酸在作物生长发育中起重要作用. 拟南芥细胞色素P450 707A3是编辑脱落酸8-羟化酶(ABA 8-hydroxylase)的基因. 我们克隆了其中的cyp707a3基因并构建了表达载体pCWori(+)-cyp707a3, 然后研究了以大肠杆菌为宿主表达CYP707A3酶的可能性. 结果表明:拟南芥细胞色素P450 707A3在E. coli. Rosseta菌株中原核表达成功并得到了具有活性的酶蛋白.实验结果都显示, 大部分酶蛋白定位于大肠杆菌细胞膜.
英文摘要:
      Abscisic acid (ABA) plays an important role in the growth and development of plants. The metabolic enzyme of abscisic acid is ABA 8-hydroxylase which are encoded by cytochrome P450 707A3 (cyp707a3) in Arabidopsis Thaliana. We cloned the cyp707a3 gene from the cDNA library and constructed the expression vector of pCWori-cyp707a3, then studied the possibility of CYP707A3 protein expression in Escherichia coli. The results showed that the prokaryotic expression was successfully in E. coli. Rosseta and active enzyme protein CYP707A3 was obtained. The experimental results showed that most of the enzyme protein was located on the cell membrane of E. coli.
