豌豆根瘤菌谷胱甘肽还原酶基因的功能研究

      谷胱甘肽还原酶将氧化型谷胱甘肽还原成还原型谷胱甘肽,是微生物体内重要的抗氧化酶类。本研究以豌豆根瘤菌RL3841的谷胱甘肽还原酶编码基因gshR为研究对象,采用基因同源重组构建豌豆根瘤菌gshR基因突变株RLgshR,并进一步研究了突变株的抗氧化和共生固氮功能。gshR基因缺失不影响菌株在AMS基础培养基中的生长能力,但突变株对氢过氧化枯(cumenehydroperoxyde,CuOOH)和较高浓度的H2O2氧化物敏感。共生实验表明, gshR基因突变株RLgshR形成正常固氮豌豆根瘤。利用实时荧光定量RT-PCR检测gshR基因的mRNA表达水平,结果显示,豌豆根瘤菌gshR基因的表达不受 H2O2和共生环境诱导。
英文摘要:
      Glutathione reductase, catalyze?the reduction of the?oxidized?glutathione to reduced?glutathione, is one of the most important bioactive antioxidants in?microorganisms. Rhizobium leguminosarum gshR mutant RLgshR was constructed through homologous recombination, and its symbiosis nitrogen fixation and antioxidative function were further studied. The growth of RLgshR was the same as the wild type RL3841 in AMS minimal medium.However, cells of the mutant were sensitive to CuOOH and H2O2 with high concentration. The symbiotic experiment showed that the gshR mutant RLgshR formed?nodules with normal?nitrogen-fixing?activity. Quantitative real-time RT-PCR was performed to detect mRNA expression of gshR gene, the results showed that gshR gene expression can not be induced under H2O2?stress?and symbiotic condition.
