      目的:本研究拟探究总黄酮是否为龙血竭阻断Kv1.3通道的主要活性组分。方法:龙血竭总黄酮经课题组优化的工艺提取成功后,采用全细胞膜片钳实验检测其对内源性和外源性表达的Kv1.3通道调节作用。结果:检测结果表明龙血竭总黄酮对内源性和外源性表达的Kv1.3通道均具有较强的抑制作用,并呈现浓度依赖性和选择性特点。结论:本研究结果提示总黄酮介导了龙血竭阻断Kv1.3通道的药理学作用,为进一步挖掘龙血竭中Kv1.3通道阻断剂提供了重要线索。
英文摘要:
      Abstract: Objective: To elucidate the main active components of Resina Draconis in blocking Kv1.3 channels. Methods: The total flavonoids of Resina Draconis (tFRD) were extracted with the optimized extraction process, and then were tested by whole cell patch clamp experiments to detect its effect on endogenous and exogenous Kv1.3 channels. Results: The tFRD had strong inhibition on endogenous and exogenous Kv1.3 channels by concentration-dependently and selectively. Conclusions: Present research not only suggested that total flavonoids mediated the pharmacological effects of Resina Draconis in blocking Kv1.3 channels, but also provided important clues for further revealing of Kv1.3 channel blockers from Resina Draconis.
