      为探讨垂盆草乙醇提取物(SSEB)对HEK293T细胞凋亡的诱导作用和机制,利用CCK-8方法检测垂盆草乙醇提取物对细胞增殖影响,应用Hoechst33342染色法及Annexin-V FITC染色流式细胞检测其对细胞凋亡影响, 应用Western blot检测其对NF-κB信号通路蛋白及凋亡相关通路蛋白BCL-2、Bax、BCL-x等表达的影响. 结果表明SSEB对细胞增殖的抑制并促进其凋亡呈剂量依赖性,Western Blotting结果表明其能显著上调NF-κΒ磷酸化水平及凋亡启动蛋白线粒体细胞色素C(cytochrome C, Cyt c)蛋白和促凋亡蛋白Bax的表达(P<0.05). 上述研究结果结果说明垂盆草乙醇提取物能诱导HEK 293T细胞的凋亡,其作用机制与NF-κΒ信号途径及其介导的凋亡启动蛋白Cyt c和促凋亡蛋白Bax的表达上调有关。

      To investigate the effect and mechanism of ethanol extract of Sedum sarmentosum (SSEB) on inducing HEK293T cells apoptosis, the cell proliferation was detected by CCK-8, cell apoptosis was detected by Hoechst33342 staining and Annexin-V FITC, and Western blotting experiments were carried out to determine the impacts of SSEB on the expression of apoptosis-related proteins, including NF-κΒ-p65, Cyt c, BCL-2, Bax and BCL-x. The results showed that the cell proliferation was significantly inhibited and the ration of cell apoptosis was significantly increased when treated with different concentration (0, 10, 50 and 100μg/mL) of SSEB (P<0.05). And the impacts of SSEB display dose-dependent relationship with SSEBon inhibition of proliferation and inducing cell apopotosis. Furthermore, the expression for apoptosis-related proteins, including NF-κΒ-p65, cytochrome C and Bax, were significantly up-regulated (P<0.05). The results indicated that SSEB can induce apoptosis of HEK 293T, and which is associated with the up-regulation of NF-κΒ signaling, Cyt c and Bax for starting and promoting apoptosis.
