      为进一步研究基因PTPMT1在肺癌细胞中的功能,我们利用CRISPR/Cas9系统建立了基因PTPMT1敲减的肺腺癌人类肺泡基底上皮细胞(A549)细胞株,并对基因敲减细胞株进行了一系列表型研究.结果表明,在A549细胞中基因PTPMT1的减少会导致细胞生长减缓.在非同步化的条件下,敲减细胞株和野生型细胞株的细胞周期并无明显变化,但在敲减细胞中细胞周期相关蛋白p27的mRNA水平显著升高.

      To investigate the function of the gene PTPMT1 in lung cancer cells, we established a lung adenocarcinoma human alveolar basal epithelial cell (A549) cell line, in which gene PTPMT1 was knockdown by the CRISPR/Cas9 system. A series of phenotypic studies were conducted on the knockdown cell lines. The results indicate that the decrease of PTPMT1 gene express in A549 cells leads to decrease of cell proliferation. Under non-synchronized conditions, compared to the wild-type cell line, the cell cycle of knockdown cell lines did not change significantly, but the mRNA level of cell cycle-associated protein P27 was significantly increased in the knockdown cells.
