      以土壤为材料,经过初筛平板富集培养,分离纯化得到能正常生长的28株细菌和35株放线菌;再分别进行液体发酵,用改进的硫酸-咔唑法测定胞内、外酶活,筛选出一株产胞外葡萄糖异构酶活力最高的细菌株菌XJ-27,酶活力为242.1U·mL-1。经对该菌株进行形态特征和16SrDNA序列比对分析,确定该菌株属于芽孢杆菌属( Bacillus)。最后对该株菌的胞外葡萄糖异构酶进行了部分性质研究,70℃测得最适pH为7.0~7.8;在pH7.4时其最适反应温度为70℃~80℃;Mg2+和Co2+对其酶活力有显著提高,Mg2+的效果最明显。
英文摘要:
      After initial screening plate enrichment culture,Twenty-eight?strains of bacteria and 35 strains of actinomycetes were isolated and purified from Multiple soil samples. Liquid fermentation was carried out separately.Then the enzyme activity was determined by improved carbazole-sulfate method. XJ-27, a bacterial strain with the highest activity of extracellular glucose isomerase was selected.The enzyme activity was 242.1 U· mL-1. XJ-27 was determined to be belonged to Bacillus by morphological characteristics and the phylogenet- ical analysis of the 16S rDNA sequence. At last, some properties of the extracellular glucose isomerase were studied. The optimum pH was 7.0-7.8.When pH7.4 its optimum reaction temperature was 70-80℃, Mg2+ and Co2+ significantly increased the enzyme activity, Mg2+ was the most effective.
