      为了提高茶多酚中活性最强的多酚物质表没食子儿茶素没食子酸酯(EGCG)的稳定性,增强其抗龋齿作用,设计构建一种含EGCG防龋齿磷脂纳米囊泡,利用纳米囊泡优良的载体性能和磷脂复合物解决EGCG稳定性问题,延长活性成分在牙齿表面吸附时间,增强EGCG的防龋齿疗效。采用紫外光谱扫描和差式扫描量热表征磷脂复合物中EGCG与磷脂的相互作用,测定纳米囊泡的粒径、多分散系数(PDI)、包封率以及形貌评价纳米囊泡的纳米特征,最后以变异链球菌评价其抗致龋菌的效果。结果表明:所得EGCG在卵磷脂的脂肪酸链的包裹下,形成EGCG磷脂复合物,以此为基础构建的EGCG纳米囊泡粒径及PDI分别为119.36 nm和0.098,包封率达到81.24±0.18%,具有良好的稳定性,对变异链球菌具有较好的抑制作用,有望在口腔防龋领域中得到应用。

      This study focuses on fabrication of phospholipid nanovesicles for encapsulation Epigallocatechin gallate (EGCG) to improve its stability, which was expected to increase the application in the Control of Oral Anti-carie. The phospholipid complex was characterized by Ultraviolet spectral scanning and Differential scanning calorimetry to explore the possibility of any interaction between EGCG and lecithin. EGCG-loaded nanovesicle was characterized for particle size, polydispersity coefficient (PDI), morphology and drug entrapment efficiency. Finally, the anti-caries activity of formulation was estimated to ensure its efficacy. The results showed that EGCG-loaded nanovesicles based on EGCG phospholipid complex had good stability and better inhibitory effect on Streptococcus mutans. The particle size and PDI of EGCG-loaded nanovesicles were 119.36 nm and 0.098, respectively, and the encapsulation rate of EGCG-loaded nanovesiclea was 81.24 ± 0.18%. It was expected to be used in the field of human oral caries prevention.
