滇重楼茎组织内生细菌的分离、鉴定及促生菌筛选

      为了开发利用重楼内生细菌资源,本研究从滇重楼茎组织中分离获得128株内生细菌,采用16S rDNA序列分析,将其鉴定归类到3个门,4个纲,6个目,11个科,14个属,其中芽孢杆菌属是优势属(75.0%),初步揭示其茎组织内生细菌的多样性. 进一步将128株内生细菌分别与小麦幼苗进行共培养,通过检测株高、根长、根数和鲜重等指标,筛选获得四株促生菌,分别是芽孢杆菌YNS21026和YNS32034、假单胞菌YNS21022和类芽孢杆菌YNS11005. 其中芽孢杆菌YNS21026对小麦的株高和根长促进效果最好,增长率分别是87.2%和310%. 该结果为后期利用促生菌改良重楼种植技术提供了菌种资源.

      In order to development and utilization of endophytic bacteria of Paris polyphylla, 128 endophytic bacteria were isolated from P. polyphylla var.yunnanensis stems and subsequently were classified into 3 phyla, 4 classes, 6 orders, 11 families, 14 genera based on 16S rDNA sequence analysis, and among them, Bacillus was one dominant genus (75.0%), which preliminary uncovered the diversity of endophytic bacteria in the P. polyphylla var.yunnanensis stems. All of the 128 endophytic bacteria were further co-cultured with wheat seedlings, respectively. Four growth-promoting endophytic bacteria were obtained through testing plant height, root length, roots number, and fresh weight of the cocultured wheat seedlings, which were Bacillus sp. YNS21026 and YNS32034, Pseudomonas sp. YNS21022 and Paenibacillus sp. YNS11005. The strain of Bacillus sp. YNS21026 had an obviously effect on the growth of plant height and root length of wheat seedlings, and the growth rates of which were promoted with 87.2% and 310%, respectively. These results provided microbial resources for improvement the cultivation technology of P. polyphylla var.yunnanensis using growth-promoting bacteria.
