      从药用植物中分离筛选能够产生活性物质的内生菌,已经成为天然产物研究的一个重要方向。本研究从白及中分离内生细菌,采用滤纸片扩散法进行抗菌活性筛选,最终得到内生细菌BSR2010能够对四种致病菌(金黄色葡萄球菌、大肠杆菌、藤黄微球菌、胡萝卜果胶杆菌)有不同程度的抑制作用;通过三种有机溶液(乙酸乙酯、石油醚、正丁醇)对发酵产物进行萃取并检测其抑菌活性,其中抑菌活性最强的为乙酸乙酯部位;根据细菌16S rDNA系统进化分析结果显示内生细菌BSR2010为假单胞菌属(Pseudomonas sp.);基因检测结果显示BSR2010菌株基因组中具有2,4-二乙酰基藤黄酚(Phl)抑菌活性物质合成的关键基因。本研究为开发利用白及内生菌资源和筛选活性化合物奠定了基础。

      The screening of endophytes that can generate active substance from medicinal plants has become an important research direction in the field of natural product. In this study, endophytic bacteria were isolated from Bletilla striata. The filter paper diffusion method was performed to screen the endophytes which had antibacterial activities. Finally,an endophytic bacterial (BSR2010) was screened which exhibited antibacterial activity of different levels against four pathogenic bacteria strains, including Staphylococcus aureus, Escherichia coli, Micrococcus luteus,Pectobacterium carolovorum. Three organic solutions (ethyl acetate, mineral- ether, n-butanol) were used to extract the fermentation product and test antibacterial activity, of which the strongest antibacterial substances were existed in ethyl acetate phase. The strain BSR2010 was identified as Pseudomonas sp. according to 16S rDNA sequence analysis. The detection results of antimicrobial metabolite synthesis genes showed that the genome of BSR2010 contained Phl related gene. This study laid the foundation for development of endophytes resources from Bletilla striata and screening active compounds.
