      目的:为研究千里光石油醚提取物(PEESS)对小鼠气管平滑肌(ASM)的舒张作用及机理.方法:使用80 mmol/L KCl或100 μmol/L Ach刺激小鼠离体气管环、小鼠气管平滑肌细胞(ASMCs)、肺部支气管以及在体小鼠气管,进行了气管环张力、细胞内Ca2+浓度、气管环面积及小鼠呼吸系统阻力检测.结果:PEESS可剂量依赖性地抑制高钾和ACh诱导的气管环预收缩,同时扩张气管环面积,降低小鼠呼吸系统阻力,这种作用是通过抑制LVDCC及NSCCs等离子通道,阻断细胞外Ca2+内流,从而舒张ASM.结论:千里光中的有效成分是作为气管扩张剂治疗哮喘的潜在药物.

      Objective: To study the relaxation effect and mechanism of petroleum ether extract from Senecio scandens(PEESS) on the airway smooth muscle (ASM) of mice. Methods: 80 mmol/L KCl or 100 μmol/L ACh were used to stimulate the isolated tracheal ring, mouse airway smooth muscle cells (ASMCs), lung bronchus and mouse trachea in vivo. The tracheal ring tension, intracellular Ca2+ concentration, tracheal ring area and respiratory system resistance in mice were then measured. Results: PEESS could dose-dependently inhibit high K+ and Ach-induced tracheal ring pre-contraction. Meanwhile, it could expand the area of tracheal ring and reduce the resistance of the respiratory system of mice. This effect was probably achieved by inhibiting the LVDCC and NSCCs channels, blocking extracellular Ca2+ influx, thereby relaxing ASM. Conclusion: The active ingredient in Senecio scandens may be a potential drug used as a tracheal dilator to treat asthma.
