      目的:探究马铃薯内生真菌Aspergillus carneus提取物中丁烯内酯类化合物LXLC-2对乳腺癌细胞(MCF-7)的凋亡诱导作用及其分子机制。方法:利用MTT法检测化合物LXLC-2对细胞活力的影响,用流式细胞术考察其对细胞凋亡的诱导,利用计算机辅助对接模拟,从配体-蛋白的自由能出发计算化合物LXLC-2与相关蛋白的对接分数,用Western blot法验证其对Caspase凋亡信号转导通路中相关蛋白表达的影响。结果:化合物LXLC-2剂量依赖性地促进MCF-7细胞凋亡,分子对接结果表明其能够与PARP、Caspase-9、P53、Bcl-2、Bax等凋亡相关蛋白结合,Western Blot实验表明化合物LXLC-2能显著下调pro Caspase-9、pro Caspase-3、PARP蛋白的表达,同时上调P53蛋白的表达。结论:化合物LXLC-2能够诱导乳腺癌细胞发生凋亡,其作用机制与Caspase级联反应以及肿瘤抑制因子P53蛋白有关。

      Objective: To explore the tumor cell apoptosis-inducing effect and mechanism of butene lactone compound LXLC-2, extracting from potato endophytic fungus Aspergillus carneus. Methods: MTT method was used to detect the effect of LXLC-2 on cell viability, and flow cytometry was used to investigate its inductive effect on cell apoptosis. Using computer-aided docking simulation, starting from the free energy of the ligand-protein, calculating the docking score of LXLC-2. Western blot was used to verify its effect on the expression of proteins related to caspase cascade. Results: LXLC-2 promoted apoptosis of MCF-7 cell in a dose-dependent manner. The molecular docking results showed that LXLC-2 can bind to PARP, Caspase-9, P53, Bcl-2, Bax and other apoptosis-related proteins. Western Blot experiment showed that LXLC-2 can significantly down-regulate the expression of pro Caspase-9, pro Caspase-3 and PARP proteins, meanwhile up-regulate the expression of P53 protein. Conclusion: Compound LXLC-2 can induce apoptosis of MCF-7 cells, and its molecular mechanism is closely related to caspase cascade and tumor suppressor P53 protein.
